Either you make the Cintiq the main display, or you configure touch in Windows.

Which tablet do you have and which pen did you buy? If your old pen works fine then the tablet and the driver are fine. The issue is only the new pen.

Your post is a little confusing but from what I understand, the issues might not be related to the driver but to the USB connection. I'm afraid that the pen sensor of your Cintiq Pro 16 has a dead scan line. The device will need to be repaired or exchanged. You should contact our support here.

Doesn't seems to be the firmware of the tablet (the pen has no firmware). As you seen in your video, the pressure indicator is consistent with the pressure you apply. The issue seems to be between in Windows Ink. Have you tried rolling the driver back to a previous version?
